Laundry room and office could be swapped, although then you would have to do puzzles in the laundry room or move ironing to the upper floor. The program is called Revit. We are currently facing the same problem with nice sliding doors - my wife also doesn't want glass and no pre-wall either -.-

As you originally set it up with your 1st post, the bathroom is above your home office; that's why I swapped the rooms. If the room layout remained like that, there would be a boxed-in area in your office, because the bathroom above would have to be drained somehow

Nowadays there are glass doors made entirely of glass. Possibly without frieze, meaning without a frame or satin-finished.
Personally, I would avoid doors, as you are also considering. But with some children? ... Or was it only one child at your place?

I will gladly inquire, but I am not aware that any measures are required for the drainage in the room below. I assumed that it goes through pipes in floors and walls...

the heating efficiency, do you think it could be neglected?

Without a door, nothing is neglected at all, unless you leave the door open for ages. For you, living only really begins around the corner - from the perspective of the front door. I must admit: we also have no door and in the super insulated house, which in winter is only ventilated with [Kontrollierte-Wohnraumlüftung], a short cold breeze is sometimes quite nice
