I'll just overlook the part about the granny flat for now. I would try to ensure you have space in the NW to maneuver the cars as well. Also, leave a bit more space in the west for the house and an evening terrace.
The slope: it's in the northeast? So it would make sense to create an outdoor access in the basement there (where the fitness room is now) and/or a (more expensive) living space/guest room/office...
I would basically move away from the rectangle and simply sketch into the plot where the turning possibility would need to be. This area could also be used multifunctionally as an evening terrace. Then maybe consider creating an inset entrance in the NW that would make it possible to do without a NW corner of the house on the ground floor, to the advantage of the turning circle. It would be nice to make the evening terrace accessible from the kitchen. Activity rooms are better suited where there are neighbors. In undisturbed corners you can chill. So I see rather relocating the entrance to the north side, kitchen in the west, living room in the east... stairs could be anywhere. Since you’re open to bay windows or offsets, you could play with two offset building structures, e.g. a crossbar with the right ridge, then a subordinate ridge parallel to the plot. Basically a big T. Or an L. I’d probably go for a stylish narrow design overall, like the barn construction.
